Overview and History
CYP4F2 encodes a member of the cytochrome P450 family (CYP4F subfamily), a class of enzymes that catalyze oxidative modifications of lipids and fat‑soluble vitamins. Biochemically, CYP4F2 performs ω‑hydroxylation reactions on substrates such as phylloquinone (vitamin K1) and certain long‑chain fatty acids, contributing to their metabolic clearance and to formation of signaling lipids.
The single nucleotide polymorphism rs2108622 (commonly reported as V433M) is a well‑studied coding variant that alters enzyme function. It was identified in pharmacogenomic and cardiovascular studies because it can change vitamin K handling and levels of bioactive fatty‑acid metabolites, producing measurable effects on biomarkers and on variability in drug response in population studies.
Interpreting Highs and Lows
Interpreting rs2108622 is framed as predicted enzyme activity rather than a circulating concentration. A genotype predicting lower CYP4F2 activity (presence of the variant allele) has been associated with higher vitamin K availability and altered production of certain vascular signaling lipids in some studies; this pattern can correlate with differences in anticoagulant dosing and with variability in vascular biomarkers. A genotype predicting typical or higher CYP4F2 activity (wild‑type) is associated with more rapid metabolism of vitamin K and related lipids.
