Overview and History
EDN1 – rs5370 is a single‑nucleotide polymorphism (SNP) in the EDN1 gene, which encodes the peptide endothelin‑1 produced mainly by endothelial cells. The SNP results in an amino‑acid change that has been studied for its effects on endothelin‑1 expression and activity.
Researchers first examined rs5370 in candidate‑gene studies of blood pressure and vascular traits; many population studies report associations between the variant and differences in vascular reactivity, endothelial biomarkers, or cardiovascular outcomes. Biologically, endothelin‑1 is a strong vasoconstrictor and modulates vascular tone, cell growth, and local inflammatory signaling, so genetic variation can influence those pathways at the molecular level.
Interpreting Highs and Lows
Higher endothelin‑1 activity or genotypes associated with increased endothelin signaling may align with patterns of greater vascular tone, altered endothelial function, or related biomarker changes (for example, differences in nitric‑oxide pathway markers or measures of vascular stress). Conversely, genotypes or measured low endothelin‑1 levels may be linked to relatively lower vasoconstrictive signaling or different vascular responses.
