Overview and History
PON1 – rs662 is a single nucleotide polymorphism (SNP) in the coding region of the PON1 gene, commonly described as the Q192R amino‑acid change. It is a genetic variant rather than a blood biomarker; different alleles at rs662 produce enzyme forms with distinct substrate preferences and catalytic rates.
Historically, rs662 drew attention in studies of both cardiovascular biology and toxicology because paraoxonase‑1 (PON1) is an HDL‑associated enzyme that hydrolyzes oxidized lipids and some organophosphates. The variant does not diagnose conditions but provides biologic context about inherited differences in enzyme function and how HDL may protect lipids from oxidative modification.
Interpreting Highs and Lows
Interpretation of rs662 centers on genotype-associated enzyme activity rather than a simple “high” or “low” concentration. One allele at rs662 is often linked to greater hydrolysis of certain substrates, while the alternate allele shows lower activity for those same substrates; effects are substrate‑specific. Patterns of lower PON1 activity (by genotype or measured activity) can suggest reduced HDL‑linked antioxidant capacity and greater tendency toward lipid oxidation, whereas higher activity patterns suggest stronger hydrolytic capacity for particular substrates. These signals are meaningful only when integrated with oxidized lipid markers, HDL function, inflammatory markers, and other Cardio Zoomer results.
