Overview and History
4-Hydroxyphenyllactic acid (4-HPLA) is an aromatic organic acid formed from the metabolism of tyrosine via host enzymatic routes and microbial transformation in the gut. Biochemically it sits within the aromatic amino acid catabolite family and is commonly measured in urine organic acid profiles as a readout of tyrosine handling.
First identified in metabolic and microbial studies, 4-HPLA has clinical research relevance as a marker of metabolic flux through aromatic pathways and of gut microbial activity. Its core role is as an end-product excreted when those pathways are active, so urinary levels reflect the balance of host enzymes, downstream metabolism, and microbiome contributions.
Interpreting Highs and Lows
Elevated urinary 4-HPLA may indicate increased tyrosine breakdown, greater microbial production of aromatic metabolites, or shifts in downstream enzyme activity; it often appears alongside other organic acids that clarify whether the source is host metabolism or microbial fermentation. Low or lower-than-expected 4-HPLA can reflect reduced substrate availability, alternative metabolic routing, or a microbial community that produces different metabolites. Either direction is nonspecific on its own and is most informative when interpreted with the full Cellular Zoomer panel (other organic acids, microbial markers, redox and detox indicators, and genetics).
