Overview and History
8‑Nitroguanine is a nitrated derivative of the nucleobase guanine, classified as a nucleic acid modification produced by reactive nitrogen species such as peroxynitrite and nitrogen dioxide. It was identified in studies of nitrative chemistry and inflammation as a specific marker of nitrogen-centered oxidative reactions on DNA and RNA.
Biologically, 8‑nitroguanine is a damage product rather than a normal cellular building block. It can form within DNA and RNA strands or as free nucleotides, and damaged bases are released and ultimately excreted. Laboratory measurement typically detects the modified base in urine or tissue extracts as an indirect indicator of ongoing nitrative processes.
Interpreting Highs and Lows
Elevated 8‑nitroguanine may indicate increased formation of reactive nitrogen species and a higher burden of nitrative stress, which can be associated with inflammation, immune activation, environmental exposures, or metabolic strain. Low levels may reflect lower nitrative activity, effective repair and clearance of damaged bases, differences in sample timing, or technical factors affecting measurement. Interpretation is most meaningful when reviewed alongside other Cellular Zoomer markers—such as DNA oxidation markers, lipid peroxidation products, antioxidant genetic variants, and mitochondrial function tests—to reveal broader redox patterns.
