Dityrosine is a covalent cross-link between two tyrosine residues created by radical-mediated chemistry (for example peroxyl radicals, peroxynitrite, or metal-catalyzed oxidation). When incorporated into proteins, these cross-links change structure and can impair normal protein function, leaving a chemically stable residue that can be measured in biological fluids. Identified in biochemical studies of oxidative protein damage, dityrosine is detected using techniques such as HPLC, mass spectrometry, or fluorescence-based assays. Because it is relatively long-lived compared with some reactive intermediates, dityrosine serves as a cumulative marker of past protein oxidative events rather than an instantaneous flux measurement.
