Overview and History

Leptin is classified as an adipokine—a signaling peptide secreted by adipose (fat) tissue. First identified in the mid-1990s from the ob gene, leptin is part of a family of hormones that communicate nutritional status to other organs. It circulates in the blood and binds receptors in the brain and peripheral tissues. Biologically, leptin’s core role is to convey information about energy stores to the brain’s appetite-regulating centers, helping adjust hunger and energy expenditure. Beyond appetite, leptin influences reproductive hormones, immune responses, and bone metabolism. While circulating leptin generally correlates with fat mass, the strength of downstream signaling varies among people, so levels alone do not capture full physiological effect.

Interpreting Highs and Lows

Higher leptin levels are often associated with greater fat mass and signals that the body has higher stored energy; they can also occur alongside chronic metabolic stress and inflammatory signaling. In some people, high leptin coincides with reduced sensitivity to its effects, a pattern sometimes described as decreased leptin signaling. Lower leptin levels are commonly seen with reduced fat stores or prolonged low-calorie states and are associated with signals that conserve energy, including reduced reproductive hormone activity and altered immune function.
