Total CK is an enzymatic biomarker: a family of creatine kinase isoenzymes (commonly CK‑MM, CK‑MB, CK‑BB) found in skeletal muscle, heart, brain, and other tissues. Clinically measurable as total CK activity in serum, it has been used for decades to monitor tissue integrity and cellular energy handling. Biochemically, CK catalyzes the reversible transfer of a phosphate group between creatine and ATP, acting as a rapid energy buffer during fluctuating cellular demand. When cell membranes are stressed or disrupted, CK can leak into the bloodstream, making serum activity a practical marker of recent muscle or cellular strain rather than a direct measure of function at the tissue level.
