Overview and History

PYD is a pyridinium crosslink that stabilizes mature collagen fibers, especially in bone and other connective tissues. Biochemically it is a non-reducible crosslink formed during collagen maturation; when collagen is degraded, PYD is released and largely excreted unchanged in urine. Clinically, PYD was identified as a specific marker of collagen breakdown and has been used as an index of bone resorption and matrix remodeling. Because PYD derives from mature collagen, its level reflects the rate of structural matrix turnover rather than short-lived metabolic fragments.
