About Centromere protein A (CENP-A)
Overview and History
CENP-A is a histone H3 variant that replaces conventional histone H3 in nucleosomes at the centromere, creating a unique chromatin signature required for kinetochore assembly and faithful chromosome segregation during cell division. As an epigenetic marker, CENP-A helps define centromere identity across cell cycles and is highly conserved among eukaryotes.
CENP-A and other centromere proteins were characterized through molecular and immunological studies; centromere components were later recognized as targets of autoantibodies in clinical serology research. Biochemically, CENP-A integrates structural and signaling roles at the centromere, recruiting protein complexes that link DNA to the mitotic spindle.
Interpreting Highs and Lows
Elevated anti-CENP-A autoantibody levels suggest measurable immune targeting of centromeric proteins and are best viewed as one element of a broader autoantibody pattern that can involve vascular or connective tissue–related markers. Low or undetectable anti-CENP-A levels indicate no measurable autoantibody reactivity to this specific centromeric protein on the assay, but do not rule out immune activity directed at other targets. Interpretation should always be done in the context of the full Immune Zoomer panel, clinical history, and symptom pattern, since isolated values have limited predictive power.
