Overview and History
GPIIb (integrin αIIb) is a membrane glycoprotein that forms a complex with β3 to create the primary fibrinogen-binding receptor on platelets. Biochemically it is classified as an integrin adhesion receptor; it mediates platelet aggregation by linking platelets to fibrinogen and other matrix proteins.
Historically, platelet glycoproteins including GPIIb were identified as major surface antigens important for platelet function and as targets of immune recognition. As an accessible surface protein, GPIIb can become an antigenic focus for antibodies or show altered surface expression, which can modify platelet behavior and vascular immune signaling.
Interpreting Highs and Lows
Elevated immune reactivity against GPIIb (higher autoantibody signal) may be associated with increased immune targeting of platelets, which can correlate with changes in platelet clearance, altered aggregation responses, or signals of vascular immune activation. Low or absent antibody reactivity generally suggests little evidence of immune targeting of this antigen. When assays measure antigen expression rather than antibodies, low antigen levels can reflect reduced platelet surface expression or lower platelet numbers, while higher antigen exposure can reflect increased platelet activation or shedding. All results should be interpreted alongside platelet counts, clinical findings, and the full Immune Zoomer panel to identify meaningful patterns.
